Abstract

The association between nuclear DNA content and ploidy level in seven populations of Tagetes spp. was determined. DNA content was estimated by flow cytometry using Zea mays L. as internal standard. Chromosome counting was performed in cytological preparations stained with acetocarmine. T. erecta, T. foetidissima , two ecotypes of T. tenuifolia and two unidentified wild populations showed a DNA average content of 2.61±0.23 pg, which corresponds to ploidy level 2x=24. DNA content variation in these diploids was 20 % among populations and 4 to 13 % within populations. DNA average content in T. patula was 5.35±0.13 pg with a ploidy level 4x=48. These results confirm that DNA average content can be used to determine ploidy levels in Tagetes.
